Immunofluorescent Staining of Heparan Sulfate in Amyloid Plaques of Alzheimer's Disease

Heparan sulfate, a glycosaminoglycan, is present in the amyloid plaques associated with Alzheimer's disease.

To visualize heparan sulfate, begin with a diseased human hippocampal tissue section containing amyloid plaques.

Fix the tissue section using a fixative to preserve cellular structures.

Next, treat the tissue with a blocking agent to prevent non-specific antibody binding.

Add a non-ionic detergent to permeabilize the cellular membranes.

Incubate the tissue with primary antibodies that specifically target heparan sulfate within the amyloid plaques.

Wash off unbound primary antibodies.

Then, add fluorophore-conjugated secondary antibodies, which bind to the primary antibodies attached to heparan sulfate.

Remove any unbound antibodies.

Stain the nuclei using a fluorescent dye.

Finally, use a fluorescence microscope to visualize the fluorescently labeled heparan sulfate within the amyloid plaques and the blue colored nuclei in the diseased tissue section.
