This study investigates the effects of antibiotics on E. coli cell division and DNA content. By using flow cytometry, the research demonstrates how antibiotics can alter bacterial physiology.
Assessing antibiotic-induced filamentation in E. coli provides a mechanistic readout for cell division inhibition, a key mode of action for certain antibacterial compounds. This dual-assay approach combining plating for viability and flow cytometry for morphological and genomic profiling enables early de-risking of antibiotic candidates by distinguishing bacteriostatic from bactericidal effects and revealing unintended impacts on DNA replication. The method supports target validation and phenotypic screening in antimicrobial discovery by linking phenotypic changes to underlying physiological disruptions.
The method fits within the antimicrobial discovery continuum from target validation through lead optimization, where morphological and genomic profiling informs compound selection and progression decisions.
