Micro-dissection of rat brain into various regions is extremely important for the study of different neurodegenerative diseases. This video demonstrates micro-dissection of four major brain regions include olfactory bulb, frontal cortex, striatum and hippocampus in fresh rat brain tissue. Useful tips for quick removal of respective regions to avoid RNA and protein degradation of the tissue are given.
Hi, I am Quint from Department of Anatomy, university of Hong Kong. Today I'm going to introduce how to micro dissect red brain into four major paths, A factor photo cortex s stratum at hippocampus. Here is a schematic image to show how we dissect the brain.
There is three cuts. The first cut plane is at the connection of of factor bulb and the frontal cortex. The second cut plane is at the ion of the corpus Callum.
The third cut plane is at an anterior tip of the phonics. After three cuts, there will be four brain paths. On the left side is a factor.
On the right side is the hippocampus. Frontal cortex lies between the first cut and the second cut. Coding nuclease, which is part of the stratum, lies between the second and the third cut on the fresh brain hemisphere.
It's very important to recognize the copa callosum and the phonics first. Then we can easily locate the three cuts. The first cut plane is at a connection of a factor bulb and the frontal cortex.
The second cut plane is at a ion of the corpus Callum, and the third cut plane is at the anterior tip of the phonics. We are going to dissect the brain and the Microscope. The dissection tools are kept ice code.
After rinsing this ice code DEPC water, we put a brain onto the ice code metal plate by half it into two Eco hemisphere, Separate or factor above from frontal cortex, collected and stored in liquid Nitrogen. Next, we start To collect frontal cortex. Cut around the anterior tip of the corpus callosum, Cut the tric cortex collected and stored it in liquid nitrogen.
Next, we dissect the straw item. Make a cut at the plane of anterior, of phonic and the anterior commission fiber, separate the coating nucleus, collect it, And store in liquid nitrogen. Finally, we are going to dissect the hippocampus.
First, we need to get rid of the midbrain so that we can have a good exposure of hippocampus. Separate the hippocampus collected and stored in liquid nitrogen, Different brain regions associated with different neurodegenerative diseases. For example, stratum relates with Parkinson's disease while frontal cortex and hippocampus relates with Alzheimer's disease.
Precise micro dissection of the brain allows us to detect mRNA and protein changes in the region.
