This article demonstrates a quantitative method to investigate mast cell exocytosis by tracking the release of secretory granules containing the fluorescent reporter protein NPY-mRFP. The protocol utilizes a calcium ionophore to induce exocytosis and employs fluorescence measurements to assess the extent of granule release.
Quantitative measurement of mast cell exocytosis using NPY-mRFP release provides a robust platform for interrogating secretory pathway mechanisms in immune cell biology. This fluorescence-based assay enables high-confidence assessment of granule release dynamics, supporting predictive evaluation of target engagement and pathway modulation in early discovery. The approach is directly relevant for de-risking immunological targets and optimizing assay systems for translational research portfolios.
This fluorescence-based exocytosis assay integrates into the discovery-to-preclinical continuum, supporting both early mechanistic studies and downstream screening workflows.
