This study investigates the cytotoxic effects of chimeric antigen receptor T cells (CAR T cells) on tumor spheroids. By utilizing fluorescent probes, the research aims to visualize apoptosis in cancer cells following CAR T cell treatment.
This assay enables quantitative assessment of CAR T cell cytotoxicity in a physiologically relevant 3D tumor model, supporting target validation and mechanistic de-risking in immuno-oncology programs. By measuring apoptosis via Annexin-V red and tracking green fluorescence loss, it provides predictive confidence in CAR T cell functional activity before in vivo studies. The method supports early discovery decisions by linking antigen engagement to apoptotic output in a scalable, imaging-compatible format.
The assay fits within the discovery continuum from lead identification (CAR construct screening) to preclinical validation, providing a functional apoptosis readout that informs go/no-go decisions based on target-dependent killing.
