BioMEMS: Forging New Collaborations Between Biologists and Engineers

This video describes the fabrication and use of a microfluidic device to culture central nervous system (CNS) neurons. This device is compatible with live-cell optical microscopy (DIC and phase contrast), as well as confocal and two photon microscopy approaches. This method uses precision-molded polymer parts to create miniature multi-compartment cell culture with fluidic isolation. The compartments are made of tiny channels with dimensions that are large enough to culture neurons in well-controlled fluidic microenvironments. Neurons can be cultured for 2-3 weeks within the device, after which they can be fixed and stained for immunocytochemistry. Axonal and somal compartments can be maintained fluidically isolated from each other by using a small hydrostatic pressure difference; this feature can be used to localize soluble insults to one compartment for up to 20 h after each medium change. Fluidic isolation enables collection of pure axonal fraction and biochemical analysis by PCR. The microfluidic device provides a highly adaptable platform for neuroscience research and may find applications in modeling CNS injury and neurodegeneration.

My Name is Newly John and I'm at Department of Biomedical Engineering at University of California Irvine. My research involves developing novel biomedical devices and microfluidic devices for neuroscience and stem cell research. Bios And micro foot is a relatively young discipline that has developed over the last maybe five to 10 years, and it uses a lot of the techniques and expertise developed for the integrated circuit industry. So what we are doing is using the miniaturization methods and approaches developed for integrated circuits to manufacture small devices with small channels and reservoirs, and use that for cell biology applications. Traditionally, most of the biological experiments have been performed on a Petri dish scale. So for the cells, when looking at a cell, which is about 10 microns in diameter, you know, basically we have a small cell that's immersed in a big volume of liquid.Using microfluidic devices, we can control the environments around the cell. The cell cell communication, as well as cell soluble factor influence much more precisely than just doing the experiment on a larger scale in a Petri dish. So the, these new developments in micro fluidic brings a lo

BioMEMS and Cellular Biology: Perspectives and Applications

Micro-scale Engineering for Cell Biology

In the past many in vitro culture systems -- mainly monolayer cultures -- often suffered from the disadvantage that differentiated primary cells had a relatively short life-span and de-differentiated during culture. As a consequence, most of their organ-specific functions were lost rapidly. Thus, in order to reproduce better conditions for these cells in vitro, modifications and adaptations have been made to conventional monolayer cultures.

Interview: Bioreactors and Surfaced-Modified 3D-Scaffolds for Stem Cell Research

This article describes a biofabrication approach: deposition of stimuli-responsive polysaccharides in the presence of biased electrodes to create biocompatible films which can be functionalized with cells or proteins. We demonstrate a bench-top strategy for the generation of the films as well as their basic uses for creating interactive biofunctionalized surfaces for lab-on-a-chip applications.

Bridging the Bio-Electronic Interface with Biofabrication

DNA origami is a powerful method for fabricating precise nanoscale objects by programming the self-assembly of DNA molecules. Here, we describe how DNA origami can be utilized to design a robotic robot capable of sensing biological cues and responding by shape shifting, subsequently relayed to a desired effect.

Designing a Bio-responsive Robot from DNA Origami

This paper presents a series of protocols for developing engineered cells and functionalized surfaces that enable synthetically engineered E. coli to control and manipulate programmable material surfaces.

Using Synthetic Biology to Engineer Living Cells That Interface with Programmable Materials

This article describes a method of transforming a low-cost commercial 3D printer into a bacterial 3D printer that can facilitate printing of patterned biofilms. All necessary aspects of preparing the bioprinter and bio-ink are described, as well as verification methods to assess the formation of biofilms.

Three-dimensional Patterning of Engineered Biofilms with a Do-it-yourself Bioprinter

Here we present a protocol to micropattern cells at single-cell resolution using DNA-programmed adhesion. This protocol uses a benchtop photolithography platform to create patterns of DNA oligonucleotides on a glass slide and then labels cell membranes with commercially available complementary oligonucleotides. Hybridization of the oligos results in programmed cell adhesion.
