كثير من البروتينات بها مواقع متعددة حيث يقترن عندها الليجند, بالإضافة إل الموقع المسؤول عن وظيفة البروتينات ربط جزيء لأي من هذه المواقع غالبًا ما ينتج عنه تغييرات تشكُلية تبدّل شكل البروتين. عندما يؤثر هذا التغيير على ربط ليجاند أخر يوصف الموقعين كمتلائمين أو متلازمين إرتباط الموقع المتلازمة يكون منظم بطريقة إيجابيًة أو سلبيًة. في الترابط الإيجابي ربط أحد الليجاند يؤدي الى تغييرات فى التشكٌل بطريقة تجعل موقع آخر أكثر احتمالاًفي الربط بالليجاند الخاص به.في الترابط السلبى،ربط أحد اليجاند يؤدي الى تغيير فى التشّكل وهذا من شأنه أن يمنع أي موقع ربط آخر من الإقتران بالليجند الخاص به. البروتينات التي تحتوي على مواقع ربط متلازم تُسمى بروتينات تفارغية وممكن أن تكون من أنواع مختلفة،مثل الإنزيمات،المستقبلات البروتينات الهيكلية،وبروتينات الحركية على سبيل المثال،اسبارتات

Allosteric proteins have more than one ligand binding site. A ligand is a molecule that binds to a protein. When a ligand binds at one site, it can affect binding at the other sites. These sites are called coupled or linked because they influence one another.

In enzymes, the site that binds the substrate is the active site. The other site is the regulatory site. When a ligand binds to the regulatory site, it can change the protein’s shape. This conformational change can then alter how the active site works.

Linkage between binding sites can be positive or negative. In positive linkage, both ligands prefer the same protein conformation. Binding at one site then increases the affinity, or binding strength, of the other site for its ligand. In negative linkage, the ligands prefer different conformations. Binding of one ligand makes it harder for the second ligand to bind to the protein.

EXPLANATION: Discussions of DNA in earlier grades have been limited to discussions of genes and the role of genes in the appearance and activities of an organism. Biology 1 represents a student's introduction to a molecular model of a DNA as well as the organization of DNA into genes and genes subsequently into chromosomes. Students should address interactions between genes in proteins which regulate both the shape and reproduction of DNA molecules. It is important to note that not all segments of DNA are transcribed for proteins. Portions of DNA are involved in regulating the expression of genes or affect the structure of the chromosomes (replication origin, centromeres, and telomeres).
