Structural Protein Function

Structural proteins are a class of proteins that support cell shape and movement. Outside of the cell, these proteins are present in structural body parts, such as bones, cartilage, and skin.&#160;
Collagen, the most abundant protein in mammals, is a structural protein found throughout the body in the extracellular matrix.&#160;
This protein winds into a triple helix that polymerizes into elongated fibrils, which then further assemble into larger fibers. Among its many structural functions, it contributes to cell and tissue shape.
The cytoskeleton is predominantly composed of structural proteins and forms the basic framework of a cell. It contains three main types of filaments: microfilaments, intermediate filaments, and microtubules.
Actin is a structural protein that makes up the microfilaments. This protein self-polymerizes into filaments and contributes to cell shape, organization, movement, and division.
Intermediate filaments can be composed of different proteins depending on the cell type. For example, keratin intermediate filaments are found in epithelial cells, whereas peripherin intermediate filaments are found in the peripheral neurons. The primary structural function of these filaments is to reinforce cells and organize them into tissues.&#160;
The microtubules consist of a class of structural proteins called tubulins. Tubulins self-assemble to form microtubules that contribute to the organization of the cytoplasm including the location of the organelles.&#160;
Structural proteins are necessary for the health of an organism, and mutations in the genes that code for these proteins can result in various diseases, including skeletal and cardiovascular abnormalities.

In connective tissue, such as skin, ligaments, and tendons, it provides tensile strength and elasticity.&#160; In bones and teeth, it mineralizes to form hard tissues and contributes to their load-bearing capacity. In addition to structural support, collagen can also interact with cell surface receptors and other intermediate molecules to regulate cellular processes, such as growth and migration, which involve changes in cell and tissue shape.
Structural proteins form the basic framework of the cell cytoskeleton.&#160; The cytoskeleton is made up of three types of filaments, microfilaments, intermediate filaments, and microtubules, and each is composed of different structural proteins. The microfilament is formed when actin self-polymerizes into long repetitive structures. These actin filaments contribute to cell shape and organization; additionally, microfilaments can also contribute to cell movement and division, when it acts in conjunction with myosin. The composition of intermediate filaments varies based on cell type. There are around 70 different genes that code for various intermediate filaments. Intermediate filaments in epithelial cells contain keratin, peripheral neurons contain peripherin, and the sarcomere in muscle cells contains desmin. The primary structural function of these filaments is to reinforce cells and organize them into tissues. Microtubules are made up of structural proteins called tubulins. Tubulins self-assemble to form microtubules that contribute to the organization of the cytoplasm, including the location of the organelles.&#160; Microtubules are also essential for mitosis and cell division.
As structural proteins are widespread, a mutation in a gene that codes for any of these proteins can have severe detrimental effects. For example, a mutation in a gene coding for collagen can result in a condition known as osteogenesis imperfecta, which is characterized by weak bones and deformities in connective tissues. Different mutations in a collagen gene can result in Alport syndrome, which is characterized by problems in organs such as kidneys, eyes, and ears.

Structural proteins give cells and tissues shape, support, and strength. They help build bones, cartilage, hair, and muscles. Important examples include collagen, actin, myosin, and keratin.

Collagen is the most abundant protein in mammals. It is found throughout the body. In skin, ligaments, and tendons, collagen adds tensile strength and elasticity. In bones and teeth, it helps form hard, load-bearing tissue through mineralization.

Collagen also does more than provide support. It can interact with cell surface receptors and other molecules to help control cellular processes. These processes include growth and migration, which involve changes in cell and tissue shape.

Structural proteins also form the cell cytoskeleton, which is the internal framework of the cell. The cytoskeleton contains three kinds of filaments: microfilaments, intermediate filaments, and microtubules. Each filament type is built from different structural proteins.

Microfilaments form when actin self-polymerizes into long, repeating strands. These actin filaments help maintain cell shape and organization. They also work with myosin to support cell movement and cell division.

Intermediate filaments vary by cell type. Around 70 different genes code for these proteins. Epithelial cells contain keratin, peripheral neurons contain peripherin, and muscle cell sarcomeres contain desmin. Their main role is to reinforce cells and organize them into tissues.

Microtubules are made of tubulin proteins. Tubulins self-assemble into structures that help organize the cytoplasm and position organelles. Microtubules are also essential for mitosis and cell division. Because structural proteins are so widespread, mutations in their genes can cause serious effects. For example, collagen gene mutations can lead to osteogenesis imperfecta, which causes weak bones and connective tissue deformities, or Alport syndrome, which affects the kidneys, eyes, and ears.
