Kinetochore-microtubule binding helps chromosomes line up and separate during mitosis. After the nuclear envelope breaks down, condensed chromosomes are exposed to the bipolar microtubules of the mitotic spindle. The kinetochore is a large, disc-shaped protein complex at the centromere region of each sister chromatid, and it serves as the microtubule binding site.

Usually, the plus-end of a single microtubule inserts into the kinetochore. In some cases, a kinetochore first makes lateral contact with the side wall of a microtubule. It then moves along that microtubule with the help of motor proteins and later forms a stable head-on attachment with the microtubule plus-end.

Early in this process, chromosomes may show a monotelic attachment. In this case, only one sister kinetochore is attached to a spindle pole, and the other sister kinetochore is still not attached to microtubules. Next, the free sister kinetochore can connect to a microtubule from the opposite spindle. This creates an amphitelic attachment, which is needed for accurate chromosome segregation.

Incorrect kinetochore attachments can also form. A syntelic attachment occurs when both sister kinetochores attach to microtubules from the same spindle pole. A merotelic attachment happens when microtubules from opposite poles bind to the same kinetochore. These errors can lead to segregation problems and can be corrected by Aurora-B kinase-dependent mechanisms.

Once the correct head-on attachment is made, more microtubules from the same spindle can join the kinetochore. This forms a kinetochore fiber that may contain about 10 to 40 microtubules in animal cells. Correct binding also creates tension as the spindle pulls while sister-chromatid cohesion resists. That tension increases microtubule-binding affinity and locks the attachment in place, helping sister chromatids achieve biorientation.
