Heart Coherence

Heart coherence is a physiological pattern in which beat-to-beat heart rate intervals become more orderly and rhythmic, often in association with coordinated autonomic regulation. In behavioral science, it matters because heart dynamics can provide a measurable window into how breathing, emotions, and stress influence attention, self-regulation, and social behavior. The pattern arises when paced breathing and emotional state shape interactions between sympathetic and parasympathetic activity, producing characteristic changes in heart rate variability that can be tracked with electrocardiography or pulse sensors. Researchers apply heart coherence measures and biofeedback to investigate stress responses and behavioral regulation, while evaluating how physiological signals may support noninvasive approaches to training attention and emotional control.

Here, the experimental protocols are described for preparing Drosophila at different developmental stages and performing longitudinal optical imaging of Drosophila heartbeats using a custom optical coherence microscopy (OCM) system. The cardiac morphological and dynamical changes can be quantitatively characterized by analyzing the heart structural and functional parameters from OCM images.

Optical coherence tomography (OCT) is a high resolution imaging technique that allows analysis of tissue specific optical properties providing the means for tissue differentiation. We developed needle based OCT, providing real-time imaging combined with on-the-spot tumor differentiation. This publication describes a method for percutaneous, needle based OCT of renal masses.

Developmental studies in the mouse are hampered by the inaccessibility of the embryo during gestation. To promote the long-term culture of the embryonic heart at late stages of gestation, we developed a protocol in which the excised heart is cultured in a semi-solid, dilute Matrigel.
