Glutamine Deamidation

Glutamine deamidation is a chemical modification in which the amide group of glutamine converts to a carboxyl group, producing glutamate and releasing ammonia. In proteins and peptides, water-mediated hydrolysis of the glutamine side chain is influenced by factors such as pH, temperature, solvent exposure, and the surrounding amino acid sequence or structure. This reaction can alter molecular charge, conformation, activity, and stability, making it important in biochemistry and protein science. Monitoring glutamine deamidation supports protein characterization, quality control of biopharmaceuticals, and the study of how chemical modifications affect biological function over time.

Here we present a step-by-step protocol of the long-length electrostatic repulsion-hydrophilic interaction chromatography-tandem mass spectrometry (LERLIC-MS/MS) method. This is a novel methodology that enables for the first time quantification and characterization of the glutamine and asparagine deamidation isoforms by shotgun proteomics.

This article will demonstrate how to monitor glutamine dynamics in live cells using FRET. Genetically encoded sensors allow real-time monitoring of biological molecules at a subcellular resolution. Experimental design, technical details of the experimental settings, and considerations for post-experimental analyses will be discussed for genetically encoded glutamine sensors.

Alcohol misuse impairs alveolar macrophage (AM) immunity due to suppressed mitochondrial respiration and bioenergetics. We recently demonstrated that ethanol (EtOH) exposure increases glutamine dependency for mitochondrial respiration in AMs. Herein, methods are provided to determine the usage of glutamine for mitochondrial respiration in EtOH-treated AMs using an extracellular flux bioanalyzer.

This video demonstrates an in vitro technique for estimating amino acid uptake by bone marrow-derived stromal cells. Na+-dependent amino acid transporters mediate the uptake of radiolabeled amino acids inside the cells, and the cellular uptake is estimated by measuring the radioactivity.

LC-MS/MS Analysis of the Proteomic Profile of Neuromelanin Granules Isolated from Human Brain Tissue

Source: Wulf, M., et.al. Laser Microdissection-Based Protocol for the LC-MS/MS Analysis of the Proteomic Profile of Neuromelanin Granules. J. Vis. Exp. (2021).This video demonstrates the analysis of peptide samples from neuromelanin granules (NMGs) isolated from human postmortem brain tissue. The peptides are first separated using high-performance liquid chromatography (HPLC) and then analyzed by mass spectrometry to determine their mass-to-charge ratios (m/z). Targeted peptides are fragmented...
