Rpe-choroid Isolation

RPE-choroid isolation is a tissue-separation technique used to obtain the retinal pigment epithelium (RPE) together with the underlying choroid for biochemical and physiological analysis. The procedure typically involves careful removal of the neural retina from an eye cup, leaving the RPE attached to the choroidal tissue through cellular and extracellular matrix interactions; controlled mechanical dissection helps preserve this interface and its molecular constituents. Isolated RPE-choroid preparations support measurements of proteins, lipids, enzymes, gene expression, oxidative stress, and barrier-related functions, providing a practical model for studying retinal metabolism, vascular interactions, and diseases affecting the outer retina.

The human retina is composed of functionally and molecularly distinct regions, including the fovea, macula, and peripheral retina. Here, we describe a method using punch biopsies and manual removal of tissue layers from a human eye to dissect and collect these distinct retinal regions for downstream proteomic analysis.
