Automated Synthesis

Automated synthesis is the use of programmable instruments to carry out chemical or biochemical reactions with limited manual intervention, improving consistency and throughput in research and manufacturing. These systems coordinate reagent dispensing, mixing, reaction timing, temperature control, and sometimes purification or in-process monitoring according to a defined workflow. In bioengineering, automated synthesis supports the preparation of peptides, nucleic acids, biomaterials, and other engineered molecules for assay development, drug research, diagnostics, and tissue engineering. By reducing operator variability and enabling parallel experimentation, it helps researchers test designs systematically and scale promising processes toward reproducible production.

The Ugi reaction has proved to be a convenient way to quickly create diverse libraries of compounds. It involves the reaction of an amine, an aldehyde, a carboxylic acid and an isonitrile typically in methanol at room temperature. In this video, we utilize a 48-slot Mettler-Toledo MiniBlock equipped with filtration tubes and a Mettler-Toledo MiniMapper automated liquid handler was used to deliver the reagents and solvent. The parameters of interest were the concentration, the solvent...

Source: Vy M. Dong and Diane Le, Department of Chemistry, University of California, Irvine, CA Merrifield's solid-phase synthesis is a Nobel Prize winning invention where a reactant molecule is bound on a solid support and undergoes successive chemical reactions to form a desired compound. When the molecules are bound to a solid support, excess reagents and byproducts can be removed by washing away the impurities, while the target compound remains bound to the resin. Specifically, we will...

Source: Amber N. Barron, Ashlea Patterson, and Taylor D. Sparks, Department of Materials Science and Engineering, The University of Utah, Salt Lake City, UT Hydrogels are a versatile class of cross-linked polymers produced through relatively simple procedures and with generally inexpensive materials. They can be formed from solution and involve a polymer backbone formed from monomer reagents, an initiator which makes the polymer reactive and a crosslinking species which binds the polymer chains...
