Cardiac Metabolism Monitoring

Cardiac metabolism monitoring is the measurement of how heart tissue produces and uses energy, providing insight into myocardial function and metabolic health. It can assess substrate utilization, oxygen consumption, glucose and fatty acid oxidation, and related changes in ATP production using physiological measurements, imaging, or biochemical assays. In bioengineering, these approaches help characterize how cardiomyocytes respond to workload, ischemia, hypoxia, drugs, and engineered tissue environments. The resulting data support studies of heart disease, metabolic remodeling, and cardiac energy efficiency, while informing the design of biosensors, organ-on-chip platforms, and therapeutic strategies that target impaired myocardial metabolism.

We describe the protocol to perform a cardiac stress test induced by dobutamine and monitored by cardiac catheterization in normal mice. Also we show its application to unmask subclinical cardiac disease in high fat diet-induced obese mice.

Here we introduce a method for using an intra-ventricle optical catheter in perfused hearts to perform absorbance spectroscopy across the heart wall. The data obtained provides robust information on tissue oxygen tension as well as substrate utilization and membrane potential simultaneously with cardiac performance measures in this ubiquitous preparation.

Metabolic Profile Analysis of Zebrafish Embryos

Zebrafish represent a powerful vertebrate model that has been under-utilised for metabolic studies. Here we describe a rapid way to measure the in vivo metabolic profile of developing zebrafish that allows the comparison of different mitochondrial function parameters between genetically or pharmacologically manipulated embryos, thereby increasing the applicability of this organism.
