Inverse Flash Nanoprecipitation

Inverse Flash Nanoprecipitation is a rapid-mixing technique for producing polymeric nanoparticles, particularly for encapsulating hydrophilic biomolecules and therapeutics. It rapidly combines an aqueous stream containing the cargo with an organic stream containing amphiphilic block copolymers, causing solvent exchange and polymer self-assembly around aqueous domains to form core-shell particles. In bioengineering, the method supports the formulation of nanoparticles containing proteins, nucleic acids, and other water-soluble agents, while controlled mixing and composition can influence particle size, loading, and stability. These properties make it useful for drug delivery, biomolecule protection, and the development of engineered nanoscale systems.

Useful tips for using the cryostat are given, including troubleshooting methods that can be used while cutting to ensure that the resultant tissues sections are free of cracks and other distortions.

Photolysis of caged compounds allows the production of rapid and localized increases in the concentration of various physiologically active compounds. Here, we show how to obtain patch-clamp recordings combined with photolysis of caged cAMP or caged Ca for the study of olfactory transduction in dissociated mouse olfactory sensory neurons.
