Multilayered Epithelium

Multilayered epithelium, also called stratified epithelium, is a tissue composed of multiple layers of closely packed cells that protects underlying structures and forms selective barriers at body surfaces. In bioengineering models, proliferating basal cells divide near a supporting substrate, while their descendants migrate outward, differentiate, and form specialized surface layers; cell junctions, extracellular matrix interactions, and controlled exposure to nutrients or air help establish tissue organization. Engineered multilayered epithelia are used to study barrier function, wound repair, disease processes, and responses to drugs or biomaterials. These systems also support the development of skin, airway, intestinal, and other tissue substitutes for regenerative medicine and testing.

Multilayer microfluidic devices often involve the fabrication of master molds with complex geometries for functionality. This article presents a complete protocol for multi-step photolithography with valves and variable height features tunable to any application. As a demonstration, we fabricate a microfluidic droplet generator capable of producing hydrogel beads.

Cells play an instrumental and increasing role in research, and the discovery and development of new therapeutics. With this increasing need for greater number of cells we need more efficient and effective ways for growing and harvesting attachment dependent cells. A Multilayered flask with the right features can serve this purpose.
