Optical Chopper Modulation

Optical chopper modulation is a measurement technique that periodically interrupts a light beam to encode a steady optical signal at a known reference frequency, improving detection of weak signals. A rotating slotted disk alternately blocks and transmits the beam, while a photodetector and lock-in amplifier isolate the response at the chopping frequency and reject much of the unmodulated background noise. In bioengineering, this approach supports sensitive optical measurements such as absorbance, fluorescence, and photometric assays, helping distinguish analyte-dependent responses from ambient light, detector offsets, and low-frequency drift. It is valuable for characterizing biosensors and optical instruments where signal-to-noise ratio and quantitative reliability are critical.

Optical biosensor techniques can detect changes in mass near the plasma membrane in living cells and allow one to follow cellular responses in both individual cells and populations of cells. This protocol will describe detection of the modulation of potassium channels by G-protein coupled receptors in intact cells using this approach.

Optically activated perfluorocarbon nanodroplets show promise in imaging applications outside of the vascular system. This article will demonstrate how to synthesize these particles, crosslink polyacrylamide phantoms, and modulate the droplets acoustically to enhance their signal.
