Photoreceptor Degenerated Retina

A photoreceptor-degenerated retina is retinal tissue in which light-sensing rod and cone cells have been lost or impaired, reducing visual signal formation and providing an important model for studying vision loss. Normally, photoreceptors convert light into electrical signals that are processed by downstream retinal neurons; degeneration disrupts this phototransduction pathway and alters retinal circuitry, while some inner retinal cells may remain available for intervention. In bioengineering, this condition guides the development of retinal prostheses, tissue-engineered scaffolds, stem-cell-derived photoreceptors, and laboratory models for testing therapies. These approaches aim to restore signaling, evaluate neural interfaces, and clarify how damaged retinal tissue can recover function.

Imaging Ca2+ Dynamics in Cone Photoreceptor Axon Terminals of the Mouse Retina

We describe a protocol to monitor Ca2+ dynamics in the axon terminals of cone photoreceptors using an ex-vivo slice preparation of the mouse retina. This protocol allows comprehensive studies of cone Ca2+ signaling in an important mammalian model system, the mouse.

The Tomato/GFP-FLP/FRT method involves visualizing mosaic photoreceptor cells in living Drosophila. It can be used to follow individual photoreceptor cell fates in the retina for days or weeks. This method is ideal for studies of retinal degeneration and neurodegenerative diseases or photoreceptor cell development.

Vibratome Sectioning Mouse Retina to Prepare Photoreceptor Cultures

Neural retina of a mouse aged 8 days is on top of a 4% gelatin block. After isolation of the photoreceptor layer (200 µm) by vibratome, the photoreceptors are seeded after mechanical and enzymatic dissociation for culture. The photoreceptor layer can be used for molecular, biochemical analyses or transplantation.
