Three Step Differentiation

Three-step differentiation is a staged cell-culture strategy that directs progenitor or stem cells toward a specialized phenotype through three sequential developmental phases. The process works by changing culture conditions at defined intervals, typically using coordinated shifts in signaling molecules, growth factors, extracellular matrix, or media composition to activate or suppress lineage-specific programs. In bioengineering, this approach helps produce more mature and reproducible cell populations for tissue engineering, disease modeling, drug evaluation, and regenerative research. Separating differentiation into discrete steps also allows researchers to control cell fate, optimize each transition, and investigate how engineered environments influence tissue development.

Retinal pigment epithelium (RPE) replacement strategies and gene-based therapy are considered for several retinal degenerative conditions. For clinical translation, large eye animal models are required to study surgical techniques applicable in patients. Here we present a rabbit model for subretinal surgery geared towards RPE transplantation, which is versatile and cost-efficient.

Rate-Determining Steps

2020

Relating Reaction Mechanisms In a multistep reaction mechanism, one of the elementary steps progresses significantly slower than the others. This slowest step is called the rate-limiting step (or rate-determining step). A reaction cannot proceed faster than its slowest step, and hence, the rate-determining step limits the overall reaction rate. The concept of rate-determining step can be understood from the analogy of a 4-lane freeway with a short-stretch of traffic-bottleneck caused due to...
