Dextran Leakage

Dextran leakage is the movement of dextran molecules from a confined fluid compartment across a permeable barrier, making it a useful indicator of barrier integrity and transport. In bioengineering studies, fluorescently labeled dextran is introduced into a vascular or microfluidic system, where its passage reflects diffusion and convection through endothelial layers or disrupted cell-cell junctions; molecular size influences the measured permeability. Researchers use this assay to evaluate vascular permeability, test engineered tissue barriers, and assess how drugs, inflammation, or physical conditions alter endothelial function. Quantifying dextran accumulation helps link barrier structure to transport behavior in disease models and tissue-engineered systems.
