Ultraviolet Visible Spectroscopy

Ultraviolet-visible (UV-Vis) spectroscopy is an analytical technique that measures how strongly a sample absorbs ultraviolet and visible light, providing information about its composition and concentration. A spectrophotometer passes selected wavelengths through the sample and records absorbance, which relates to concentration through the Beer-Lambert law; changes in absorbance can also indicate reaction progress or molecular interactions. In bioengineering, UV-Vis spectroscopy supports protein and nucleic acid quantification, enzyme kinetics, microbial growth measurements, and monitoring of bioprocesses. Its rapid, non-destructive measurements make it useful for evaluating biomaterials, optimizing laboratory protocols, and tracking biological production systems.

Source: Laboratory of Dr. B. Jill Venton - University of Virginia Ultraviolet-visible (UV-Vis) spectroscopy is one of the most popular analytical techniques because it is very versatile and able to detect nearly every molecule. With UV-Vis spectroscopy, the UV-Vis light is passed through a sample and the transmittance of light by a sample is measured. From the transmittance (T), the absorbance can be calculated as A=-log (T). An absorbance spectrum is obtained that shows the absorbance of a...

Ultraviolet and Visible (UV–Vis) Spectroscopy: Overview

2024

Ultraviolet–visible (UV–visible or UV–Vis) spectroscopy is an analytical technique that investigates the interaction between matter and UV–Vis light within the electromagnetic spectrum. This method is widely used for its versatility, simplicity, and relatively quick data acquisition, making it valuable for both qualitative and quantitative analysis. When UV–Vis radiation passes through a material, molecules absorb light depending on the energy required for electronic transitions. As a result...

2025

This video demonstrates the use of a UV-visible spectrophotometer for monitoring insoluble fibrin clot formation as a function of the change in the solution turbidity. The UV-visible spectrophotometer measures the solution’s absorbance over time, which represents the turbidity change due to the insoluble fibrin clot formation.

Light from astronomical objects must travel through the earth's turbulent atmosphere before it can be imaged by ground-based telescopes. To enable direct imaging at maximum theoretical angular resolution, advanced techniques such as those employed by the Robo-AO adaptive-optics system must be used.
