Acidic Alcian Blue

Acidic Alcian Blue is a histochemical staining technique that detects acidic mucins and glycosaminoglycans in biological tissues, making otherwise subtle extracellular materials visible by light microscopy. Under acidic conditions, the positively charged copper phthalocyanine dye binds negatively charged sulfate and carboxyl groups on polysaccharides, producing an intense blue signal. Researchers use the stain to identify mucus-producing cells, cartilage matrix, connective tissue components, and other carbohydrate-rich structures in tissue sections. Its staining pattern supports studies of tissue organization, secretory activity, development, and disease-related changes, including alterations in mucin production or extracellular matrix composition.

The preparation and exfoliation of CaCuSi4O10 and BaCuSi4O10 are described. Upon stirring in hot water, CaCuSi4O10 spontaneously exfoliates into monolayers, whereas BaCuSi4O10 requires ultrasonication in organic solvents. Near infrared (NIR) imaging illustrates the NIR emission properties of these materials, and aqueous dispersions of these nanomaterials are useful for solution processing.
