Floral Dipping Technique

The Floral Dipping Technique is a plant transformation method that introduces foreign DNA into reproductive tissues, enabling efficient generation of genetically modified plants without tissue culture. In the commonly used Arabidopsis system, flowering plants are immersed in an Agrobacterium suspension, often containing a surfactant, allowing the bacterium to transfer T-DNA into developing ovules or embryos. Transformed seeds are then selected and grown to establish stable transgenic lines. This approach supports gene-function studies, promoter analysis, genome engineering, and functional genomics by simplifying transformation and reducing the time, equipment, and labor required for regeneration-based methods.

This article illustrates the floral-dip method of Agrobacterium tumefaciens -mediated transformation of Arabidopsis thaliana. By introducing a cell-cycle regulated promoter-reporter, pTSO2::β-glucuronidase (GUS), into Arabidopsis, we illustrates how one detects GUS reporter expression in transgenic seedlings.

Agrobacterium-mediated transformation using a floral-dip method can be successfully employed to create stable transgenic lines of the extremophyte model Schrenkiella parvula. We present a protocol modified from that for Arabidopsis thaliana, considering different growth habits and physiological characteristics of the extremophyte.

Here, we present a protocol to transform flax using Agrobacterium-mediated plant transformation via floral-dip. This protocol is simple to perform and inexpensive, yet yields a higher transformation rate than the current available methods for flax transformation.

Here, we present the aluminum chloride colorimetric method, a direct analytical technique to determine flavonoids in calendula quantitatively. This approach utilizes a straightforward chemical reaction involving treating the calendula extract with an aluminum chloride reagent, forming a colored complex. The color intensity, assessed using spectrophotometry, correlates with the flavonoid concentration.
