Acetyl Coa Metabolism

Acetyl-CoA metabolism is the set of biochemical pathways that produces, uses, and redistributes acetyl-CoA, a central two-carbon metabolite linking carbohydrate, lipid, and amino acid metabolism. In mitochondria, pyruvate dehydrogenase converts pyruvate to acetyl-CoA, while fatty acid oxidation and selected amino acid catabolism provide additional sources; acetyl-CoA then condenses with oxaloacetate in the citric acid cycle to generate reducing equivalents and support ATP production. When energy or carbon demands change, acetyl-CoA can be exported as citrate for fatty acid synthesis or diverted to ketone body formation, making its regulation important in fasting, exercise, and metabolic disease research.

Here we show isolated human platelets can be used as an accessible ex vivo model to study metabolic adaptations in response to the complex I inhibitor rotenone. This approach employs isotopic tracing and relative quantification by liquid chromatography-mass spectrometry and can be applied to a variety of study designs.
