Acetylated Tubulin Staining

Acetylated tubulin staining is an immunofluorescence technique that detects acetylated α-tubulin, a post-translationally modified form of tubulin associated with specific microtubule populations. In a typical assay, cells are fixed and permeabilized, then incubated with a primary antibody against acetylated tubulin and a labeled secondary antibody; fluorescence microscopy reveals the distribution and organization of these microtubules. In biology, the method helps visualize stable microtubule structures, including axons, cilia, and other polarized cellular extensions. It supports studies of cytoskeletal organization, cell morphology, intracellular transport, and changes in microtubule stability during development, disease, or drug treatment.

This manuscript describes an experimental approach to morphologically and biochemically characterize horse oocytes. Specifically, the present work illustrates how to collect immature and mature horse oocytes by ultrasound-guided ovum pick-up (OPU) and how to investigate chromosome segregation, spindle morphology, global histone acetylation, and mRNA expression.

Optimizing Tubulin Yield from Porcine Brain Tissue

This protocol describes a technique for the high-yield isolation of tubulin from the porcine brain optimized for small-scale instrumentation. The isolation procedures are complemented by procedures for determining tubulin polymerization activity in vitro using co-sedimentation assays and transmission electron microscopy.
