Acf Subunits

ACF subunits are the protein components of the ATP-dependent chromatin assembly factor (ACF), a molecular complex that organizes DNA into functional chromatin. The complex combines the ISWI ATPase, which uses energy from ATP hydrolysis to reposition nucleosomes, with ACF1, which helps recognize and organize nucleosomal DNA and regulates remodeling activity. By controlling nucleosome spacing and chromatin accessibility, ACF subunits influence gene regulation, DNA replication, and genome maintenance. Studying their composition and interactions helps explain how cells package genetic material while preserving access to regulatory sequences, with implications for developmental biology, disease mechanisms, and chromatin-based research.

Here, we describe a detailed protocol for the in vitro reassociation of purified, salt-washed eukaryotic ribosomal subunits for the analysis of 80S particle formation. This method is illustrated by analyzing ribosomal subunits isolated from a wild-type strain of Saccharomyces cerevisiae and a mutant strain deficient in the ribosomal protein eL24.

This publication presents a protocol showing how to measure the activity of recombinant protein phosphatase 2A catalytic subunit (PP2Ac) in response to recombinant α-synuclein, β-synuclein, or γ-synuclein proteins using a simple colorimetric assay. This study introduces a multiscale framework, spanning from DNA to protein function and neural behavior. It presents a novel approach for investigating predicted pathogenic mutations in the GABAA receptor subunit, hypothesizing that epileptogenic mutations and proximal mutations, predicted as pathogenic, may produce similar effects on the CA1 pyramidal neuron model.
