Adipocytes

Adipocytes are specialized cells that store excess energy as triglycerides and help regulate whole-body metabolism, making them central to biology and health. They develop from precursor cells through adipogenesis, accumulate lipid droplets, and release fatty acids through lipolysis when hormonal signals such as catecholamines indicate increased energy demand. Adipocytes also function as endocrine cells, secreting adipokines that influence appetite, inflammation, insulin sensitivity, and tissue communication. Studying white, brown, and beige adipocytes helps researchers understand energy balance, thermogenesis, obesity, type 2 diabetes, and potential strategies for treating metabolic disease.

Adipose tissue (AT) can influence whole body homeostasis, therefore understanding the molecular mechanisms of adipocyte differentiation and function is of importance. We provide a protocol for gaining new insights into these processes by analyzing adipocyte homeostasis, differentiation and hypoxia exposure as a model for induced adipocyte apoptosis.

This video demonstrates the differentiation of bone marrow stromal cells (BMSCs) into adipocytes. In the two-step process, in the presence of specific growth factors and hormones, undifferentiated BMSCs first differentiate into preadipocytes. The second step involving treatment with glucose and insulin leads to the formation of mature adipocytes with multiple lipid droplets.

Mature adipocytes may represent an abundant source of stem cells through dedifferentiation, which leads to a homogenous population of fibroblast-like cells. Collagenase digestion is used to isolate mature adipocytes from human fat. The goal of our protocol is to obtain multipotent, dedifferentiated fat cells from human mature adipocytes.

Here we report a method for isolation of Adipocyte Progenitor Cell (APC) populations from Perivascular Adipose Tissue (PVAT) using Magnetic-activated Cell Sorting (MCS). This method allows for an increased isolation of APC per gram of adipose tissue when compared to Fluorescence-Activated Cell Sorting (FACS).
