Protein Topology Determination

Protein topology determination is the process of establishing how a protein is arranged within or across a biological membrane, including the number and orientation of its transmembrane segments and the locations of its termini. Researchers infer topology by combining sequence-based hydropathy analysis with experimental approaches such as protease protection, selective labeling, reporter fusions, and membrane fractionation. These methods reveal which protein regions face the cytosol, extracellular space, or organelle lumen and help distinguish membrane-spanning domains from soluble regions. Accurate topology maps support studies of protein folding, trafficking, signaling, and transporter function, while guiding the design of mutational analyses and functional models in cell biology.
