Adipogenic Osteogenic

Adipogenic and osteogenic differentiation are biological processes in which progenitor or stem cells develop into adipocytes, which store lipids, or osteoblasts, which produce bone-forming matrix. These distinct cell fates arise through lineage-specific signaling and transcriptional programs, often activated by defined culture conditions that promote lipid accumulation for adipogenesis or extracellular matrix production and mineralization for osteogenesis. Comparing these pathways helps researchers characterize cellular potential, study tissue development and remodeling, and evaluate cell-based strategies for fat and bone regeneration. Adipogenic and osteogenic assays are also useful for assessing how biological or material environments influence mesenchymal cell fate.

Quantitative measurement of bone progenitor function in fracture healing requires high resolution serial imaging technology. Here, protocols are provided for using intravital microscopy and osteo-lineage tracking to sequentially image and quantify the migration, proliferation and differentiation of endogenous osteogenic stem/progenitor cells in the process of repairing bone fracture.

Polyelectrolyte Complex for Heparin Binding Domain Osteogenic Growth Factor Delivery

Self-assembled polyelectrolyte complexes (PEC) fabricated from heparin and protamine were deposited on alginate beads to entrap and regulate the release of osteogenic growth factors. This delivery strategy enables a 20-fold reduction of BMP-2 dose in spinal fusion applications. This article illustrates the benefits and fabrication of PECs.

This protocol outlines the isolation of skeletal and cardiac muscle fibro-adipogenic progenitors (FAPs) from spiny mouse (Acomys) via enzymatic dissociation and fluorescence-activated cell sorting. The FAPs obtained from this protocol can be effectively expanded and differentiated to myofibroblasts and adipocytes.

Visualization and Quantification of Mesenchymal Cell Adipogenic Differentiation Potential with a Lineage Specific Marker

Traditional methods of assessing adipogenic differentiation are cheap and easy to use, but are not specific to changes in gene expression. We have developed an assay to quantify mesenchymal cell differentiation into mature adipocytes using a lineage specific marker. This assay has diverse applications across basic research and clinical medicine.
