Agilent Bioanalyzer Analysis

Agilent Bioanalyzer Analysis is a microfluidic method for assessing nucleic acid and protein samples by separating components and measuring their size and abundance. In a disposable chip, samples are loaded into wells, driven through microchannels by electrophoresis, and detected with fluorescence to generate electropherograms and quality metrics such as RNA integrity. In biology, this analysis helps verify RNA or DNA quality before sequencing, gene expression studies, and other workflows, while size distribution and concentration measurements can reveal degradation, contamination, or inconsistent sample preparation.

Alcohol misuse impairs alveolar macrophage (AM) immunity due to suppressed mitochondrial respiration and bioenergetics. We recently demonstrated that ethanol (EtOH) exposure increases glutamine dependency for mitochondrial respiration in AMs. Herein, methods are provided to determine the usage of glutamine for mitochondrial respiration in EtOH-treated AMs using an extracellular flux bioanalyzer.

The isolation of high quality, intact RNA is an essential step in many laboratory protocols. Here, we demonstrate RNA extraction from whole zebrafish embryos and cDNA synthesis for subsequent application in various experimental procedures including gene expression microarray analysis.
