Antibiotic Synergy Analysis

Antibiotic synergy analysis is the evaluation of whether two or more antibiotics work together to inhibit or kill bacteria more effectively than either drug alone. Researchers measure combined activity by exposing bacterial cultures to varying drug concentrations and comparing outcomes such as growth inhibition, killing, or changes in the fractional inhibitory concentration index. This approach can reveal complementary mechanisms, including disruption of different cellular targets or improved access to bacterial processes. In biology and antimicrobial research, synergy analysis helps identify promising combination therapies, optimize treatment strategies, and assess whether drug pairings may overcome reduced susceptibility or limit the development of resistance.

We describe a platform that utilizes a library of isogenic antibiotic resistant Escherichia coli for the dereplication of antibiotics. The identity of an antibiotic produced by bacteria or fungi can be deduced by the growth of E. coli expressing its respective resistance gene. This platform is economically effective and time-efficient.
