Antibody Enrichment

Antibody enrichment is a laboratory process that increases the proportion or concentration of target antibodies within a complex biological sample, improving their suitability for analysis or experimentation. The method typically uses selective affinity interactions, such as binding between an antibody and its antigen or between antibody Fc regions and an affinity ligand, to capture desired antibodies while other proteins are removed during washing. Elution then releases the enriched antibodies for downstream use. In biology, antibody enrichment supports immunoassays, protein characterization, structural studies, and antibody development by improving sample purity, detection sensitivity, and experimental consistency.

2013

We describe the use of the monoclonal antibodies TG30 (CD9) and GCTM-2 for the combined detection of cell surface antigens via fluorescence activated cell sorting (FACS) for the identification and enrichment of live human embryonic stem cells (hESC) using positive selection and also the use of negative selection to purge hESCs from a mixed cell population.

Scalable High Throughput Selection From Phage-displayed Synthetic Antibody Libraries

A method is described with visual accompaniment for conducting scalable, high throughput selections from phage-displayed combinatorial synthetic antibody libraries against hundreds of antigens simultaneously. Using this parallel approach, we have isolated antibody fragments that exhibit high affinity and specificity for diverse antigens that are functional in standard immunoassays.

2026

All animal procedures described here must be conducted in accordance with institutional animal ethics guidelines and approved by IACUC. All procedures must follow the principles of the 3Rs—Replacement, Reduction, and Refinement—and must be performed by trained personnel. Environmental enrichment involves objects and practices that encourage natural behaviors, improve psychological well-being, and promote better welfare in laboratory rodents. Environmental enrichment should be tailored to the...
