Autoimmune Encephalitis

Autoimmune encephalitis is a group of inflammatory brain disorders in which the immune system mistakenly targets neuronal proteins, disrupting brain function and potentially causing rapid, serious neurological symptoms. In many cases, antibodies bind to cell-surface receptors or ion channels, altering synaptic signaling and triggering additional immune inflammation; other forms involve immune responses against intracellular antigens. Symptoms can include memory loss, seizures, psychiatric changes, movement abnormalities, and impaired consciousness. Research and clinical evaluation combine neurological examination, antibody testing, brain imaging, cerebrospinal fluid analysis, and electroencephalography to guide immunotherapy, improve diagnosis, and clarify how immune mechanisms affect neural circuits.

Autoimmune encephalitis is a new category of antibody-mediated diseases of the central nervous system. Hippocampal neurons can be used to discover and characterize these antibodies. This article provides a protocol for primary cell culture and immunostaining to determine autoantibodies in the serum and cerebrospinal fluid of patients.
