Axon Imaging

Axon imaging is the visualization of axons, the slender neuronal processes that transmit signals from cell bodies to target cells, and is essential for studying neural connectivity and function. Using light or fluorescence microscopy, researchers label axonal structures or molecular indicators and capture their morphology, transport, growth, degeneration, or activity over time; calcium-sensitive probes can reveal changes associated with electrical signaling. In biology, axon imaging helps characterize neural development, synaptic communication, injury responses, and neurodegenerative disease. Quantitative analysis of axon length, branching, movement, and signal dynamics can connect cellular mechanisms with nervous-system organization and support evaluation of experimental treatments.

Live Imaging of Dorsal Root Axons after Rhizotomy

An in vivo imaging protocol to monitor primary sensory axons following dorsal root crush is described. The procedures utilize wide-field fluorescence microscopy and thy1-YFP transgenic mice, and permit repeated imaging of axon regeneration over 4 cm in the PNS and axon interactions with the interface of the CNS.

This study presents an in vivo two-photon imaging approach for visualizing the structures of low-threshold mechanoreceptor (LTMR) axon terminals in the forepaw skin of mice. Using repeated, high-resolution imaging of individual axons, this method provides a new platform for studying the structure and function of the cutaneous sensory circuit during development and in adults.
