Bacterial Characterization

Bacterial characterization is the systematic identification and description of bacterial traits, helping researchers distinguish organisms and understand their biological roles. It combines observations of cell shape, colony morphology, staining behavior, growth conditions, and metabolic activity with biochemical or molecular analyses, such as tests for characteristic enzymes or detection of genetic markers. In biology, these approaches can identify clinically relevant bacteria, clarify microbial diversity and interactions, and support accurate classification. Characterization also informs antibiotic selection, contamination control, environmental monitoring, and the development of useful strains for biotechnology.

We describe an efficient, robust, and cost effective method for extracting nucleic acid from swabs for characterization of bacterial communities using 16S rRNA gene amplicon sequencing. The method allows for a common processing approach for multiple sample types and accommodates a number of downstream analytic processes.

Here we present a protocol to produce gram-negative Escherichia coli (E. coli) spheroplasts and gram-positive Bacillus megaterium (B. megaterium) protoplasts to clearly visualize and rapidly characterize peptide-bacteria interactions. This provides a systematic method to define membrane localizing and translocating peptides.
