Bead Beating Homogenization

Bead beating homogenization is a mechanical cell-disruption technique that uses rapidly agitated beads to break biological samples into a uniform mixture for analysis. During processing, collisions between the beads and sample generate impact, shear, and friction that rupture cell walls or membranes, releasing nucleic acids, proteins, and other intracellular components; bead size, material, agitation speed, and processing time influence the result. In biology, this method supports sample preparation from microorganisms, plant tissues, animal tissues, and other difficult-to-lyse materials. Efficient homogenization improves extraction quality and reproducibility for downstream molecular, biochemical, and microbiological assays.

This protocol presents a technique for probing protein-protein interactions using glutathione-linked donor beads with GST-fused TPR-motif co-chaperones and acceptor beads coupled with an Hsp90-derived peptide.
