Bone Cell Isolation

Bone cell isolation is a laboratory technique used to separate viable cells from bone tissue for studying skeletal biology and disease. The process typically involves removing soft tissue, finely mincing or pulverizing bone, and applying mechanical disruption or enzymatic digestion, often with collagenase, to release cells from the mineralized matrix. Researchers can then enrich, culture, and characterize populations such as osteoblasts, osteocytes, or bone marrow-derived cells using microscopy, molecular assays, and functional tests. Isolated bone cells support investigations of bone formation and resorption, cellular responses to drugs or mechanical stimuli, and disorders including osteoporosis and bone tumors.

Murine Hind Limb Long Bone Dissection and Bone Marrow Isolation

Here we present a protocol for the dissection of hind limb long bones (femurs and tibiae) from the laboratory mouse. We further describe a rapid technique for bone marrow isolation from these bones that utilizes centrifugation for removal of bone marrow from the bone marrow space.

Here we describe an optimized, highly reproducible protocol to isolate Mesodermal Progenitor Cells (MPCs) from human bone marrow (hBM). MPCs were characterized by flow cytometry and nestin expression. They showed the ability to give rise to exponentially growing MSC-like cell cultures while retaining their angiogenic potential.

A protocol establishing mesenchymal stem cells (MSCs) isolated from the synovial fluid and bone marrow of minipigs and non-invasively collected using syringe aspiration is presented. Cellular phenotyping was performed using flow cytometry after cell isolation and in vitro cultivation of bone marrow and synovial fluids derived from MSCs.

Here we present a protocol that generates large amounts of murine monocytes from heterogeneous bone marrow for translational applications. In comparison to others, this new method helps reduce the number of sacrificed animals and lowers costs by avoiding expensive methods such as high gradient magnetic cell separation (MACS).
