Bone Structure

Bone structure is the organized arrangement of mineralized tissue, connective tissue, marrow, and living cells that gives bones strength, flexibility, and support. Its outer compact bone contains tightly packed osteons, while inner spongy bone forms a lightweight network of trabeculae; collagen fibers provide tensile resilience, and calcium phosphate minerals provide stiffness. Bone structure continually adapts through remodeling, in which osteoclasts remove old tissue and osteoblasts deposit new matrix in response to mechanical stress and physiological needs. Understanding this architecture supports the study of movement, mineral homeostasis, fracture repair, osteoporosis, skeletal development, and the design of biomaterials for orthopedic applications.

Here we present a protocol for the dissection of hind limb long bones (femurs and tibiae) from the laboratory mouse. We further describe a rapid technique for bone marrow isolation from these bones that utilizes centrifugation for removal of bone marrow from the bone marrow space.
