Box-behnken Design

Box-Behnken design is a response surface methodology for modeling how several experimental factors influence a measured response while reducing the number of runs needed. It uses three coded levels for each factor and tests combinations in which two factors vary between low and high settings while the remaining factors stay at their center levels, supplemented by replicate center points. By avoiding combinations with all factors simultaneously at extreme settings, it can estimate curvature and interactions across a practical experimental region. In biology, researchers apply Box-Behnken design to optimize culture conditions, enzyme production, extraction procedures, assay performance, and other multivariable workflows, helping identify influential factors and balanced settings for improved yield, activity, or reproducibility.
