Calcium Dependent Binding

Calcium-dependent binding is the reversible interaction of calcium ions with proteins, nucleic acids, membranes, or other molecules, allowing cells to regulate biological activity in response to calcium signals. Calcium binds specific sites through charged amino acids, often inducing a conformational change that exposes interaction surfaces or promotes molecular bridging, as seen in EF-hand and C2 domains. This mechanism helps control neurotransmitter release, muscle contraction, enzyme activation, cell adhesion, and intracellular signaling. Studying calcium-dependent binding clarifies how transient changes in calcium concentration produce coordinated cellular responses and supports research into physiology, disease mechanisms, biomaterials, and therapeutic strategies.

In this video, we demonstrate the purification of calcium-binding protein from a dialyzed cell lysate through calcium-dependent hydrophobic interaction chromatography. The calcium-binding proteins expose a hydrophobic region upon binding with calcium, facilitating interaction with a hydrophobic group on resin. Later these proteins are eluted using calcium chelator EDTA that reverses the interaction.
