Cardiomyocyte Generation

Cardiomyocyte generation is the biological process of producing heart muscle cells, or cardiomyocytes, from stem cells, progenitor cells, or other differentiated cell types. In laboratory systems, researchers guide cells through mesoderm formation and cardiac specification by regulating developmental signals, particularly the timing and intensity of Wnt pathway activation and inhibition, followed by maturation under defined culture conditions. Generated cardiomyocytes support studies of heart development, inherited and acquired cardiac disease, and drug-induced toxicity. They also provide models for testing therapies and may contribute to tissue engineering and regenerative medicine, although challenges such as cellular maturity, structural organization, and functional integration remain.

Pluripotent stem cells, either embryonic or induced pluripotent stem (iPS) cells, constitute a valuable source of human differentiated cells, including cardiomyocytes. Here, we will focus on cardiac induction of iPS cells, showing how to use them to obtain functional human cardiomyocytes through an embryoid bodies-based protocol.

We describe here a protocol for the generation of iCMs using retrovirus-mediated delivery of Gata4, Tbx5 and Mef2c in a polycistronic construct. This protocol yields a relatively homogeneous population of reprogrammed cells with improved efficiency and quality and is valuable for future studies of iCM reprogramming.

Primary mouse cardiomyocyte cultures are one of the pivotal tools for the investigation of myofibrillar organization and function. The following protocol describes the isolation and culture of primary cardiomyocytes from neonatal mouse hearts. The resulting cardiomyocyte cultures may be subsequently used for a variety of biomechanical, biochemical and cell-biological assays.

Isolation and Cryopreservation of Neonatal Rat Cardiomyocytes

The isolation of neonatal rat cardiomyocytes is a time consuming and unpredictable procedure. This study describes methods for cryopreservation and thawing of neonatal rat cardiomyocytes that allows for more efficient use of cells. The thawed NRCMs can be used for various experiments without the need for performing isolations each time.
