Cdna Purification

cDNA purification is a laboratory method that isolates complementary DNA after reverse transcription, producing a cleaner template for downstream molecular analysis. The process removes residual RNA, primers, free nucleotides, enzymes, salts, and other reaction components, commonly by binding cDNA to a silica membrane or magnetic beads under high-salt conditions, washing away contaminants, and eluting the nucleic acid in a low-salt buffer. Purified cDNA supports reliable PCR and quantitative PCR, cloning, sequencing, and transcriptome library preparation. By reducing inhibitors and background, purification improves amplification efficiency, measurement accuracy, and the reproducibility of gene expression studies.

The cDNA microarray PtGen2 was developed for gene expression studies in loblolly pine, P. taeda, and other conifer species. Here, we show pre- and post-hybridization handling and washing techniques that can be used with this array to yield better consistency, reduced artifacts, and lower backgrounds.
