Cell Clump Preparation

Cell clump preparation is a laboratory method for producing compact aggregates of cells, enabling researchers to study cellular behavior in a three-dimensional context rather than as isolated cells. The process typically involves collecting cells from a culture, concentrating them, and maintaining close cell-cell contact so adhesion and extracellular matrix interactions promote aggregate formation while preserving cell viability. Prepared clumps can support three-dimensional culture, directed differentiation, organoid development, and tissue engineering studies. By providing a more tissue-like environment, this approach helps investigate cell organization, communication, development, and responses to experimental conditions in biology.

A novel protocol for the mechanical preparation of testicular cell suspensions from rodent material, avoiding enzymes and detergents, is described. The method is very simple, fast, reproducible, and renders good quality cell suspensions, which are suitable for flow sorting and RNA extraction.
