Cell Culture Hypoxia

Cell culture hypoxia is the controlled reduction of oxygen available to cells grown in vitro, a condition used to model oxygen-limited environments found in tissues and disease. Lower oxygen tension alters mitochondrial respiration and stabilizes hypoxia-inducible factors (HIFs), which regulate gene expression involved in metabolism, angiogenesis, survival, and stress responses. Researchers create hypoxic cultures with specialized incubators, gas mixtures, or chemical oxygen-mimetic treatments, then assess cellular adaptation under defined conditions. This approach supports studies of tumor biology, stem cell maintenance, development, ischemia, and tissue engineering while helping distinguish oxygen-dependent effects from other experimental variables.

Human Primary Trophoblast Cell Culture Model to Study the Protective Effects of Melatonin Against Hypoxia/reoxygenation-induced Disruption

This manuscript presents a unique in vitro model of immunopurified human villous cytotrophoblast cells cultured under hypoxia/reoxygenation. This model is suitable to study the protective effects of promising treatments, such as melatonin, on pregnancy complications associated with increased oxidative stress and altered placental function.

Delivery of In Vivo Acute Intermittent Hypoxia in Neonatal Rodents to Prime Subventricular Zone-derived Neural Progenitor Cell Cultures

This article describes the methodology for administering short periods of intermittent hypoxia to postnatal day 1-8 mouse or rat pups. This approach effectively elicits a robust tissue level “priming effect” on cultured neural progenitor cells that are harvested within 30 min of hypoxia exposure.

Analysis of Global RNA Synthesis at the Single Cell Level following Hypoxia

We describe a technique for analysis of global RNA synthesis in hypoxia using imaging. Click-chemistry labeling of RNA has not previously been performed under hypoxia and allows visualization of global RNA changes at the single cell level. This approach complements the existing averaged RNA techniques, allowing direct visualization of cell-to-cell changes in global RNA synthesis.

Modeling Hypoxia/Reoxygenation Injury in Proximal Tubular Epithelial Cells

Here, we describe an in vitro protocol to model hypoxia/reoxygenation damage in the highly metabolic proximal tubular epithelial cells. This model is designed to induce metabolic stress-induced damage, which can be measured through the expression of proximal tubule damage markers and assessment of mitochondrial respiratory function.
