Cell Cycle Arrest

Cell cycle arrest is a regulated state in which a cell temporarily or permanently stops progressing through the cell division cycle, helping preserve genome integrity and control tissue growth. This pause occurs when signals such as DNA damage, nutrient limitation, or differentiation cues activate checkpoint pathways that inhibit cyclin-dependent kinases, preventing transitions between phases such as G1, S, and G2/M. In biology, studying cell cycle arrest clarifies how cells respond to stress, enter senescence, or coordinate development, while also informing cancer research, where defective checkpoint control can permit uncontrolled proliferation and induced arrest may support treatment strategies.

Studying Cell Cycle-regulated Gene Expression by Two Complementary Cell Synchronization Protocols

We report two cell synchronization protocols that provide a context for studying events related to specific phases of the cell cycle. We show that this approach is useful for analyzing the regulation of specific genes in an unperturbed cell cycle or upon exposure to agents affecting the cell cycle.

Determining the cell cycle position of a population of cells, or understanding how signals affect proliferation, can be readily measured by flow cytometry using this protocol. We report a simple experimental approach to staining cells and quantifying their position in the cell cycle.
