Cell Membrane Recruitment

Cell membrane recruitment is the regulated localization of proteins, lipids, or signaling molecules at a cell’s plasma membrane, where proximity enables coordinated cellular responses. It occurs when membrane-binding domains recognize specific lipids, lipid modifications increase membrane affinity, or adaptor and scaffold proteins capture soluble factors at defined membrane sites. Recruitment can activate signaling pathways, organize cytoskeletal remodeling, control vesicle trafficking, and coordinate cell adhesion or movement. In biology research, measuring membrane recruitment helps reveal how cells translate external cues into spatially controlled biochemical events and supports studies of development, immunity, disease mechanisms, and potential therapeutic targets.

This manuscript describes a method to quantify tumor cell accumulation in the lungs in an animal model of tumor metastasis.

An In Vitro Flow Investigation of the Effect of Stromal Cells on Leukocyte Recruitment

The video demonstrates a method for investigating the impact of stromal cells on leukocyte recruitment. In a parallel plate flow chamber model, pro-inflammatory cytokines induce the expression of adhesion receptors by endothelial cells, facilitating neutrophil recruitment and migration. The cross-talk between stromal cells and endothelial cells diminishes neutrophil recruitment to the endothelial cells.

The ability of inflamed endothelium to recruit leukocytes from flow is regulated by mesenchymal stromal cells. We describe two in vitro models incorporating primary human cells that can be used to assess neutrophil recruitment from flow and examine the role that mesenchymal stromal cells play in regulating this process.

Monitoring Neutrophil Recruitment Dynamics and Bacterial Burden at the Infection Site in a Murine Model

This video demonstrates in vivo imaging to study the neutrophil recruitment dynamics at an infection site in an immunocompromised, transgenic mouse with GFP-expressing neutrophils. It involves injecting luminescent bacteria to cause an infection. This leads to neutrophil recruitment to the infection site, whose tracking is done by imaging.

We describe a matrigel plug assay to illustrate angiogenic potential of a pool of factors secreted by cancer cells using two complementary imaging modalities, ultrasound and endomicroscopy. The matrigel, an extracellular matrix (ECM)-mimic gel, is utilized to introduce the host (mouse) with angiogenic factors secreted to the conditioned media...
