Collagen Solution Preparation

Collagen solution preparation is the process of dissolving and conditioning collagen to produce a uniform material for biological experiments, where its concentration, pH, and handling influence subsequent use. Typically, collagen is dispersed in an acidic aqueous medium to maintain solubility, then mixed carefully and, when a gel is required, neutralized so collagen molecules assemble into fibrils under physiological conditions. Prepared solutions support cell culture matrices, three-dimensional tissue models, wound-healing studies, and biomaterial fabrication, providing an extracellular-matrix-like environment for investigating cell adhesion, migration, and tissue organization.

Traditional procedures for the isolation of soluble type 1 collagen (COL1) require about 10 days from start to finish because of lengthy buffer incubations and laborious resuspensions of fibrils. Here, we describe a means to purify COL1 from small dermal biopsies in less than 3 hr.

Collagen is a core component of the ECM, and provides essential cues for several cellular processes ranging from migration to differentiation and proliferation. Provided here is a protocol for embedding cells within 3D collagen hydrogels, and a more advanced technique for generating randomized or aligned collagen matrices using PDMS microchannels.

2023

Collagen is another widely used biomaterial that has found popularity in commercial applications, such as photography. Collagen has more recently been used in tissue engineering applications, by creating hydrogels that provide structure to engineered tissue. This video introduces collagen as a biomaterial, demonstrates how it is harvested from porcine skin, and shows how the material is used to create a hydrogel for tissue engineering applications. Finally, several applications of the material...

Preparation of Naringenin Solution for In Vivo Application

2021

Here, the protocol presents the preparation of naringenin solution for in vivo intraperitoneal administration. Naringenin is fully dissolved in a mixture of dimethylsulfoxide, Tween 80, and saline. The antidiabetic osteoporotic effects of naringenin were assessed by blood glucose testing, tartrate-resistant acid phosphatase staining, and enzyme-linked immunosorbent assay.
