Cell Recovery

Cell recovery is the process of restoring cell viability and function after isolation, culture disruption, physical stress, or cryopreservation, allowing cells to resume normal biological activity. Following thawing or transfer, cells are placed in an appropriate growth medium and controlled environment while, when necessary, cryoprotectants or debris are removed; survival depends on membrane integrity, metabolic recovery, attachment, and adaptation to culture conditions. Reliable cell recovery supports cell culture, primary-cell studies, stem-cell research, tissue engineering, and reproducible experiments by providing healthy populations for downstream analysis, expansion, and treatment testing.

Olfactory ensheathing cells (OECs) are neural crest cells which allow growth of the primary olfactory neurons. This specific property can be used for cellular transplantation. We present here a model of cellular transplantation based on the use of OECs in a laryngeal nerve injury model.

The following protocol provides techniques for encapsulating pancreatic β-cells in step-growth PEG-peptide hydrogels formed by thiol-ene photo-click reactions. This material platform not only offers a cytocompatible microenvironment for cell encapsulation, but also permits user-controlled rapid recovery of cell structures formed within the hydrogels.

Recovery of Zika Virus from In Vitro-Transcribed RNA

Source: Weger-Lucarelli, J., et al. Rescue and Characterization of Recombinant Virus from a New World Zika Virus Infectious Clone. J. Vis. Exp. (2017)This video demonstrates the recovery of infectious Zika virus using electroporation of in vitro transcribed RNA. The process shows how synthetic RNA initiates infection and leads to viral replication in cultured cells.
