Hair Follicle Dissection

Hair follicle dissection is a laboratory technique that separates the structural components of a hair follicle for microscopic, molecular, or cellular analysis. Using fine instruments, researchers isolate regions such as the outer root sheath, dermal papilla, and hair matrix while preserving tissue architecture and cell viability when required. This approach helps reveal how epithelial and mesenchymal cells interact to regulate follicle development, cycling, and regeneration. In biology, hair follicle dissection supports studies of stem cell behavior, tissue organization, pigmentation, wound repair, and skin disorders, while providing material for culture, transplantation, and gene expression analyses.

The hair follicle is a highly complex appendage of the skin containing a multiplicity of cell types. The follicle undergoes constant cycling through the life of the organism including growth and resorption with growth dependent on specific stem cells. The targeting of the follicle by genes and stem cells to change its properties, in particular, the nature of the hair shaft is, discussed. Hair follicle delivery systems are described, such as liposomes and viral vectors for gene therapy. The...

Accessory Structures of the Skin: Hair and Hair Follicles

2025

Hair and hair follicles are integral components of the integumentary system. Hair is a filamentous structure composed mainly of a protein called keratin. It is found on the surface of the skin throughout the body, except for areas such as the palms of the hands and soles of the feet. Hair is a keratinous filament growing out of the epidermis. It is primarily made of dead, keratinized cells. Hair strands originate at the epidermal penetration called the hair follicle. The hair shaft is the part...

Combined Recording of Mechanically Stimulated Afferent Output and Nerve Terminal Labelling in Mouse Hair Follicles

This video demonstrates the combined recording of mechanically stimulated afferent output and nerve terminal labeling in the hair follicles of a mouse pinna. It outlines the steps involved in the mechanical stimulation of hair follicle nerve terminals before and after exposure to a fluorescent membrane dye, electrophysiology recording of the generated action potentials, and visualization of the nerve terminal labeling in the hair follicles.
