Cell Sheets

Cell sheets are contiguous layers of living cells that can be detached from culture surfaces while preserving cell-cell junctions and much of their extracellular matrix. They are commonly produced on temperature-responsive culture surfaces, where a controlled temperature change weakens cell adhesion to the substrate and allows the intact layer to be harvested without enzymatic dissociation. This structure supports cell-sheet stacking, transplantation, and integration with host tissues, making the approach valuable in tissue engineering and regenerative medicine. Cell sheets also provide models for studying tissue organization, wound repair, disease processes, and cell-based therapies while retaining interactions that isolated cells may lose.

For creation of highly organized structures of complex tissue, one must assemble multiple material and cell types into an integrated composite. This combinatorial design incorporates organ-specific layered cell sheets with two distinct biologically-derived materials containing a strong fibrous matrix base, and endothelial cells for enhancing new vessels formation.

In this video, we describe the lattice light-sheet microscopy technique to visualize interactions between live T-cells and antigen-presenting cells. The method involves focusing an ultrathin lattice light sheet on an interacting cell pair to obtain a series of two-dimensional images, which are combined to obtain high-resolution four-dimensional images that reveal the spatiotemporal dynamics of the cell-cell interaction.

Using Light Sheet Fluorescence Microscopy to Image Zebrafish Eye Development

Light sheet fluorescence microscopy is an excellent tool for imaging embryonic development. It allows recording of long time-lapse movies of live embryos in near physiological conditions. We demonstrate its application for imaging zebrafish eye development across wide spatio-temporal scales and present a pipeline for fusion and deconvolution of multiview datasets.
